I'm no A/C expert but I believe it's usually based on high side pressure vs low side pressure, based on the outside air temperature while the unit is running.

About 4.3 lbs R-134a. Charge until the temp in the air ducts bottoms out and stop. You'll want the duct temp to be 20-25* below the outside temp.
